چکیده

Since Schaper and his coworkers1-3 reported an excellent coronary vasodilating action of lidofiazine, a drug originally prepared in Janssen Pharmaceutica Institute, on conscious dogs with a previously implanted electromagnetic flow meter, the antianginal effect of the drug has been clinically investigated in our own and other laboratories. Its usual dose is 90 or 120mg per day. Recently, we administered a large dose of lidoflazine (90mg three times a day) to a patient with atrial fibrillation due to coronary heart disease, and a reversion to sinus rhythm was unexpectedly recognized. Since there has been no report on such a defibrillating effect of this drug, and since a chemical structure of lidofiazine is similar to that of lidocain which has been recommended for clinical use as an effective antiarrhythmic drug,4-7 we think it adequate to evaluate the effect of lidofiazine in the treatment of atrial fibrillation and of other arrhythmias. The present paper is a preliminary report of the results obtained up to the present. Patients with established atrial fibrillation were divided into two groups according to the procedure of medication. The first group consisted of seven

Comparison of lidoflazine and quindine in prophylactic treatment of arrhythmias.

The efficacy of quinidine versus lidoflazine therapy was compared in the maintenance of sinus rhythm after electrical cardioversion for atrial flutter or fibrillation in a group of 35 patients. Both quinidine and lidoflazine were relatively ineffective in maintaining sinus rhythm after cardioversion. Lidoflazine was also used to prevent supraventricular and ventricular tachycardias in a miscell...

Cardioprotective effects of lidoflazine during 1-hour normothermic global ischemia.

The cardioprotective effects of lidoflazine, a drug with calcium homeostatic properties, were investigated in dogs subjected to 1 hour of normothermic global ischemia, followed by reperfusion. None of the eight control dogs could be weaned from the extracorporeal bypass, confirming the severity of the ischemic model. All eight acutely pretreated dogs showed rapid recovery from the prolonged isc...

Role of Fructose as a Potent Antiarrhythmic and Anti-infarct agent in Isolated Rat Heart

In the current study, effects of acute short term administration of fructose on cardiac arrhythmias and myocardial infarction size following ischemia/reperfusion were investigated in isolated rat heart. The hearts were subjected to 30 min zero flow global ischemia followed by 120 min reperfusion. In the control group, the hearts were perfused by normal drug free Krebs-Henseleit (K/H) solution t...

The effect of intravenous lidoflazine on serotonin-induced cerebral vascular contraction--an in vivo study.

Lidoflazine, a piperazine derivative with known selectivity for vascular smooth muscle, was evaluated as a possible agent for prophylaxis of cerebral vascular contraction induced by subarachnoid perfusion with serotonin. The animals treated with serotonin (5 X 10(-6) M), had a 60% reduction in the diameter of basilar artery but when pretreated with Lidoflazine (1 mg/kg) intravenously, only had ...

A unitary mechanism of calcium antagonist drug action.

[3H]Nitrendipine binding to drug receptor sites associated with calcium channels is allosterically regulated by a diverse group of calcium channel antagonists. Verapamil, D-600 (methoxyverapamil), tiapamil, lidoflazine, flunarizine, cinnarizine, and prenylamine all reduce [3H]nitrendipine binding affinity. By contrast, diltiazem, a benzothiazepine calcium channel antagonist, enhances [3H]nitren...
